為 Coomassie brilliant blue G-250 法研製而成,以 Biuret 法為基礎進一步改良,藉由protein於鹼性溶液中會將二價的 Cu 離子還原為一價的銅離子,並與 BCA 形成結合,並在OD.595 nm 時.偵測吸光值,進行蛋白質定量
